Vilela,Laíze Aparecida Ferreira; Saggin Júnior,Orivaldo José; Paulino,Helder Barbosa; Siqueira,José Oswaldo; Santos,Vera Lúcia da Silva; Carneiro,Marco Aurélio Carbone.
Soil structure has major influence on ecosystem sustainability and plant growth. Arbuscular mycorrhizal fungi (AMF) are an important functional group of soil microbiota, acting in the process of aggregation, especially in agroecosystems and related to the production and plant diversity. AMF are widely distributed in tropical agroecosystems and are extremely important for development of many agricultural crops. The present study evaluated the effect of inoculation with Glomus macrocarpum in different crop sequences on plant growth, microbial activity and aggregation of a Cerrado Oxisol. The study was conducted in a completely randomized design in a4 x 4 factorial scheme. Treatments consisted of four conditions of elimination and/or introduction of AMF...

ABSTRACT In Southern Brazil, agricultural soils are often treated over long periods with annual pig slurry (PS) applications, and their influence on soil nutrient dynamics should be monitored. This study aimed to evaluate the P forms accumulated in the soil and some biological and biochemical parameters of the P cycle after 19 successive PS applications over eight years, in a no-tillage (NT) system. Pig slurry was applied at rates of 0, 20, 40, and 80 m3 ha-1 per crop. Several P forms (available P, microbial P, phosphatase enzyme, total organic and inorganic P) in the 0.00-0.05 and 0.05-0.10 m soil layers were analyzed. Increases up to 5,523 % in available P, 308 % in microbial P, 104 % in phosphatase enzyme activity, 91 % in organic P, 251 % in inorganic...

Cytochemical localization of acid phosphatase in Stigeoclonium tenue (Chaetophorales, Chlorophyceae) Biocell
Michetti,Karina M.; Leonardi,Patricia I.; Cáceres,Eduardo J..
Nonspecific acid phosphatases are a group of enzymes whose activity increases the availability of exogenous and endogenous orthophosphate either through extra- or intracellular hydrolysis of phosphate compounds. Our study demonstrates the activity of acid phosphatases in the filamentous freshwater alga Stigeoclonium tenue. These enzymes were detected following a cerium-based method in which cerium was used as an orthophosphate-capture reagent. In thalli from S. tenue from the natural environment, acid phosphatases were found in the longitudinal cell wall, plasmalemma, and vacuole. In thalli from Bold's Basal Medium culture, these enzymes were found mainly in the plasmalemma; they were scarce in the cell wall. In the thalli grown in phosphate-enriched...

Dissolving mechanism of strain P17 on insoluble phosphorus of yellow-brown soil BJM
Chuan-qing,Zhong; Guang-xiang,Cao; Wei-yi,Huang; Xing-she,Luan; Yi-fei,Yang.
Strain P17 was a bacterial strain identified as Bacillus megaterium isolated from ground accumulating phosphate rock powder. The fermentation broth of strain P17 and the yellow-brown soil from Nanjing Agricultural University garden were collected to conduct this study. The simulation of fixed insoluble phosphorous forms after applying calcium superphosphate into yellow-brown soil was performed in pots, while available P and total P of soil were extremely positive correlative with those of groundwater. Then the dissolving effect of strain P17 on insoluble P of yellow-brown soil was studied. Results showed that Bacillus megaterium strain P17 had notable solubilizing effect on insoluble phosphates formed when too much water-soluble phosphorous fertilizer...

Extracellular breakdown of collagen by mice decidual cells. A cytochemical and ultrastructural study Biocell
Godosevicius Katz,Sima.
The interaction of antimesometrial decidual cells and collagen fibrils was studied by light microscopy and ultrastructural cytochemistry in fed and acutely fasted mice on days 9-11 of pregnancy. Fibrillar elements in the extracellular space consisted of collagen fibrils and filamentous aggregates (disintegrating collagen fibrils). Intracellular vacuoles exhibited typical collagen immersed in electron-translucent material (clear vacuoles) and faint cross-banded collagen immersed in electron-opaque material (dark vacuoles). Fibrillar elements showed extracellular acid phosphatase activity which was stronger in the region of mature decidua than in predecidual cells region in all animals; it was conspicuous in mature decidua of fasted animals. Intracellular...

Lowering of phytic acid content by enhancement of phytase and acid phosphatase activities during sunflower germination BABT
Agostini,Juliana da Silva; Nogueira,Rosicler Balduíno; Ida,Elza Iouko.
The objective of this work was to investigate the germination of hybrid sunflowers BRS191 and C11 as a means of lowering phytic acid (PA) content by enhancing the activity of endogenous phytase and acid phosphatase. The concentration of PA in hybrid sunflower achenes varied from 2.16 to 2.83g/100g of sample (p < 0.05). The phytase and acid phosphatase activities of sunflowers BRS191 and C11 were the highest on the 4th and 5th days of germination, respectively, with the release of the phosphorus. These results indicated that hybrid sunflower PA reduced and enhance phytase activity at distinct germination periods, which could open up the possibility of applying these enzymes in the control of PA content in cereals, thus improving their nutritional value.

Phosphate solubilization by endophytic bacteria isolated from banana trees Anais da ABC (AABC)
MATOS,AMANDA D.M.; GOMES,IZABELA C.P.; NIETSCHE,SILVIA; XAVIER,ADELICA A.; GOMES,WELLINGTON S.; DOS SANTOS NETO,JOSÉ A.; PEREIRA,MARLON C.T..
ABSTRACT Forty isolates of endophytic bacteria isolated from banana tree roots were assessed as to their capacity to solubilize phosphate in a solid culture medium supplemented with different inorganic and one organic source of phosphorus. The amount of phosphorus (P) in each liquid medium was quantified, and an indirect assessment of acid phosphatase activity was performed. All assays had a fully randomized design, with three repetitions. Approximately 67.5% of the 40 isolates assessed in solid medium solubilized phosphorus from tricalcium phosphate and 7.5% of the isolates solubilized phosphorus from soy lecithin; no isolates exhibited P solubilization capacity in medium supplemented with iron phosphate. Acid phosphatase activity was detected in 65% of...

The lysosomal stability as a biomarker for the determination of pollution in aquatic environments BABT
Nazar,Maria Loreto; Rodrigues,Luiz Erlon Araujo; Nascimento,Iracema.
This work studied the effects caused by five different formulae of gasoline on the stability of the lysosomes isolated from the liver of the tilapia fish (Oreochromis niloticus). The functional integrity of the lysosomal membranes was evaluated via the acid phosphatase activity. The results showed that there were significant changes in the stability of the lysosomes exposed to the presence of the hydrocarbons in the environment. Therefore, considering the method's simplicity, the sensitivity of the responses and its low cost the assessment of the lysosomal activity could be an important tool for the study of the effects of pollution in the aquatic environments.
